Description

From STEMCELL Technologies, Human Recombinant IL-34 (Interleukin 34) is a dimeric glycoprotein belonging to the short-chain helical hematopoietic cytokine family. The protein carries a polyhistidine tag at the C-terminus and is purified as a homodimer consisting of 39 kDa monomers, with a predicted molecular mass of 26.7 kDa. IL-34 is known for its ability to induce the formation of colony-forming unit macrophages in human bone marrow cell cultures and exists in two isoforms that differ by a single glutamine. It interacts with M-CSF to trigger tyrosine phosphorylation of the receptor and ERK1/2 pathways, and in combination with RANKL it induces osteoclast differentiation.

Expressed in CHO cells, the product is supplied as a lyophilized powder prepared from sterile PBS at pH 7.4, with trehalose (5% - 8%), mannitol, and 0.01% TWEEN® 80 normally included as protectants before lyophilization. Biological activity is determined by a cell proliferation assay using human peripheral blood mononuclear cells (PBMCs), with an EC50 of 2 - 8 ng/mL. Purity is ≥ 92% and the endotoxin level is ≤ 1.0 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis.

For preparation, the vial should be centrifuged before opening and reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial, without vortexing. The product should be stored at -20°C to -80°C and is stable as supplied for 12 months from the date of receipt.
