HotStarTaq Plus DNA Polymerase (25000)

A hot-start DNA polymerase from QIAGEN for end-point PCR, the enzyme requires an initial heat-activation step of 5 min at 95°C before cycling begins. Because the polymerase is held in an inactive state until this activation step, non-specific DNA synthesis cannot occur at room temperature during reaction setup. The kit is supplied with PCR Buffer that provides a final concentration of 1.5 mM MgCl2 in the reaction, Q-Solution to facilitate amplification of GC-rich templates or those with complex secondary structure, and CoralLoad PCR Buffer, which contains a gel-loading reagent and gel-tracking dyes so amplification products can be loaded directly onto an agarose gel without adding a separate loading buffer. CoralLoad PCR Buffer must not be used in capillary sequencers. All components should be stored immediately upon receipt at –30 to –15°C in a constant-temperature freezer.

Manufacturer-recommended cycling conditions include denaturation at 94°C for 0.5–1 min, annealing at 50–68°C for 0.5–1 min (approximately 5°C below the primer Tm), and extension at 72°C for 1 min per kb, with a final extension of 10 min at 72°C and 25–35 cycles in total. QIAGEN has announced the discontinuation of the HotStarTaq Plus product line, with availability after November 30, 2023 limited to stocks on hand and AllTaq PCR kits recommended as the successor.
