HiSpeed Plasmid Maxi Kit (25)

An ultrafast maxi-scale workflow from QIAGEN isolates ultrapure, supercoiled plasmid DNA using a modified alkaline lysis procedure combined with gravity-flow anion-exchange chromatography with QIAGEN Resin HS. The protocol, provided with the HiSpeed Plasmid Maxi Kit, eliminates the need for ultracentrifugation, HPLC, or toxic reagents such as phenol and ethidium bromide, and yields DNA suitable for transfection, automated or manual sequencing, and enzymatic modifications in less than one hour.

The procedure starts with pelleting an overnight LB culture of 150 ml (high-copy plasmid) or 250 ml (low-copy plasmid), followed by resuspension in 10 ml Buffer P1, lysis with 10 ml Buffer P2, and neutralization with 10 ml prechilled Buffer P3. The cleared lysate is processed through a QIAfilter Maxi Cartridge without centrifugation and applied to a HiSpeed Maxi Tip operating by gravity flow. After washing with 60 ml Buffer QC, DNA is eluted in 15 ml Buffer QF and concentrated via isopropanol precipitation using the QIAprecipitator Maxi Module. The precipitated DNA is washed with 2 ml 70% ethanol and eluted in 1 ml Buffer TE.

The kit is for research use only and components are stored dry at room temperature (15–25°C), with HiSpeed Tips, QIAfilter Cartridges, and QIAprecipitator Modules remaining stable for at least two years.
