GloCell FV Dye Violet 540, 100 tests

Designed for live/dead staining of mammalian cells in flow cytometry, this amine-labeling fluorescent dye is offered by STEMCELL Technologies as GloCell™ Fixable Viability Dye Violet 540. The dye irreversibly binds intracellular and cell surface amine groups and can be used prior to fixation, permeabilization, or cryopreservation. Cells with compromised plasma membranes become permeable to the dye, resulting in greater fluorescence compared to live cells in a cell viability assay.

According to the product information sheet, the dye is excited by the violet laser at 405 nm and has a fluorescence emission maximum of 540 nm, detectable using the 525/50 band pass filter typically used for AmCyan. A spectral reference from the manufacturer lists an excitation maximum of 399 nm and an emission maximum of 540 nm.

The product is supplied in anhydrous DMSO and should be stored at -20°C, protected from prolonged exposure to light, and used before the expiry date printed on the label. The dye has been verified for Flow Cytometry (FC) and is reported for Immunofluorescence microscopy (IF); it has also been verified for viability assessment of cells obtained with EasySep™ and RosetteSep™ kits. For staining, cells are resuspended at 1 - 10 x 10^6 cells/mL in D-PBS (without Ca++ and Mg++), 1 µL of dye is added per 1 mL of cell suspension, and the suspension is incubated at 2 - 8°C for 30 minutes in the dark.
