Description

A fluorescent amine-labeling dye from STEMCELL Technologies, this fixable viability stain is designed for discriminating live and dead mammalian cells in flow cytometry and immunofluorescence applications. The dye irreversibly binds both cell surface and intracellular amine groups: live cells retain intact membranes and show only limited surface staining, whereas cells with compromised membranes permit dye entry, yielding markedly higher fluorescence that clearly separates the two populations in flow cytometry data.

Excited by the violet laser at 405 nm, the dye has a fluorescence emission maximum of 510 nm and can be detected using the 525/50 band pass filter used for AmCyan. Cells labelled with the dye retain their fluorescence intensity following fixation, permeabilization, and washing, so the stain can be applied prior to downstream fixation, permeabilization, or cryopreservation steps. The product is also compatible with intracellular antibody staining protocols and can be used for visual discrimination of viable and dead cells during fluorescence microscopy.

Supplied in anhydrous DMSO, the dye should be stored at -20°C, protected from prolonged exposure to light, and remains stable until the expiry date on the label. Typical handling involves resuspending washed cells in D-PBS at 1 - 10 x 10^6 cells/mL, adding 1 µL of dye per 1 mL of cell suspension, and incubating for 30 minutes at 2 - 8°C in the dark, followed by washes in a protein-containing buffer. Flow cytometry is a verified application, with immunofluorescence microscopy also reported; the dye has been verified for viability assessment of cells isolated with EasySep™ and RosetteSep™ kits. For research use only; not intended for human or animal diagnostic or therapeutic uses.
