Description

STEMCELL Technologies supplies this flavonoid compound, which occurs naturally in plants and vegetables and exhibits senolytic activities. The compound upregulates expression of sirtuin 1 (SIRT1), a member of the sirtuins superfamily of histone deacetylases (HDAC) that helps inhibit transcription factors including p53, nuclear factor kappa B (NF-κB), and forkhead box O (FOXO). It also inhibits DNA methyltransferase 1 (DNMT1)-mediated DNA methylation through direct inhibition of the DNA methylase, with an IC50 of 3.5 μM reported.

The compound is identified by CAS Number 528-48-3, chemical formula C15H10O6, and molecular weight of 286.2 g/mol, with a reported purity of ≥ 90%. Alternative names include CI-75620, NSC 407010, and NSC 656275, and the chemical name is 2-(3,4-dihydroxyphenyl)-3,7-dihydroxy-4H-1-benzopyran-4-one. The product is supplied as a crystalline solid and remains stable for 12 months from date of receipt when stored at -20°C away from direct light, with long-term storage recommended with a desiccant.

Solubility is reported in DMSO at ≤ 100 mM and absolute ethanol at ≤ 17 mM, with low solubility in aqueous media. The manufacturer's published applications span several research areas: maintenance and self-renewal studies showing enhanced survival of PC12 cells from tunicamycin (Tm)-mediated cell death; immunology research showing downregulation of phorbol-12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated gene expression and production of TNF-α, IL-1β, IL-4, IL-6, and IL-8 in human mast cells; cancer research demonstrating inhibition of the PI3K/Akt and mTOR pathway in prostate, lung, myeloma, and melanoma cancer cells, alongside antioxidant activity; and disease modeling research demonstrating ERK activation with neuroprotective effects in Huntington's disease models and reductions in senescence and age-related pathologies in yeast, simple eukaryotes, and mice.
