EZ2 RNA/miRNA Tissue/Cells Kit (48)

QIAGEN offers an automated solution for isolating total RNA, including miRNA and mRNA, from human and animal cells and tissues. The kit is designed for use with the EZ2 Connect instrument family and uses magnetic-particle technology. Purified RNA is suitable for downstream applications such as RT-PCR, digital PCR (dPCR), and next-generation sequencing (NGS), with protein and genomic DNA removed during the process.

The workflow begins with manual lysis in Buffer RLT, followed by Proteinase K digestion to ensure complete lysis of difficult tissues such as muscle or brain. After a 10 minute incubation at room temperature, samples and prefilled reagent cartridges are loaded onto the EZ2 Connect, where bind, wash, and elute steps are fully automated. An on-instrument DNase step digests contaminating genomic DNA, after which the RNA is rebound to the magnetic beads and washed to remove residual contaminants.

The kit accepts up to 30 mg of frozen tissue or 15 mg of stabilized tissue (or 10 mg frozen spleen or lung tissue, or up to 5 x 10^6 cells), with RNA eluted in either 50 or 100 µl. The protocol uses no phenols and is compatible with all versions of the EZ2 Connect, including the EZ2 Connect, EZ2 Connect Fx, and EZ2 Connect MDx.
