EZ2 AllPrep DNA/RNA FFPE Kit (48)

The QIAGEN EZ2 AllPrep DNA/RNA FFPE Kit enables automated and simultaneous purification of DNA and RNA from formalin-fixed, paraffin-embedded (FFPE) tissue samples. Used with the EZ2 Connect instrument, the kit automates extraction of both nucleic acid types from the same piece of sample in two distinct elution fractions, using magnetic-bead technology and prefilled cartridges.

The kit technology allows reversal of formalin-induced crosslinking without overnight proteinase K incubation. In the patented workflow, FFPE tissue samples are incubated in a lysis buffer that releases RNA into the supernatant while leaving DNA in the pellet. The DNA-containing pellet and RNA-containing supernatant are separated by centrifugation, then processed in separate tubes within the same automated run using a single cartridge. This approach permits up to 24 samples per run while allowing optimal crosslink removal conditions for each nucleic acid type, since RNA is more susceptible to heat-induced fragmentation than DNA but is also less affected by crosslinks.

The bind, wash, and elute steps are fully automated, shortening the workflow to only a few manual steps. Specified elution volumes are RNA: 50–60 µl and DNA: 100–120 µl, with a maximum sample amount of 4×10 µm sections or 2×20 µm sections. The kit is automated-compatible and intended for use with EZ2 automation platforms, while also being adaptable to manual workflows. Purified DNA and RNA are suitable for real-time PCR, (RT-)qPCR, NGS, genotyping, and microarray analysis.
