EZ1&2 DNA FFPE Kit

From QIAGEN, this kit enables automated purification of genomic DNA from formalin-fixed, paraffin-embedded (FFPE) tissue samples on the EZ1 Advanced XL or EZ2 Connect instrument. It uses silica-based magnetic-bead technology with a two-step Proteinase K lysis procedure, a wash-free, no-solvent deparaffinization technique, and a crosslink-removal step to maximize recovery of amplifiable DNA from difficult-to-lyse samples.

FFPE tissue sections of 5–10 µm thickness, totaling 4 mm³, can be processed, with elution volumes of 60 µl or 100 µl. The purified genomic DNA is suitable for PCR, digital PCR, and next-generation sequencing workflows, and may be stored at −30 to −15°C. An optional uracil-N-glycosylase (UNG) workflow removes deaminated cytosine residues, reducing artifactual C→T/G→A transitions and limiting false-positive single-nucleotide variant calls during NGS analysis. On the EZ2 Connect, up to 24 samples can be processed in a single run, with deparaffinization, binding, washing, and elution automated; manual preparation steps such as Proteinase K digestions, crosslink removal, and RNase A digestion are performed off-instrument.

Buffers and reagent cartridges are shipped at ambient temperature and stored at 15–25°C; cartridges must not be frozen. The ready-to-use Proteinase K solution is stable for at least 1 year when stored appropriately. The kit is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
