Description

Anion-exchange technology underpins QIAGEN's EndoFree Plasmid Giga Kit, delivering up to 10 mg of endotoxin-free, advanced transfection-grade plasmid or cosmid DNA from 2.5 liters of culture [S2, S3]. The manual, gravity-flow workflow processes 1 sample per run in 285 minutes and accommodates high-copy, low-copy, and cosmid plasmid types [S2, S3].

Bacterial lysates are cleared by filtration through QIAfilter Mega-Giga Cartridges, after which plasmid DNA binds to the resin in gravity-flow QIAGEN-tips [S2, S3]. An integrated endotoxin-removal step removes the need for additional extractions or affinity columns, yielding DNA with endotoxin levels below 0.04 EU/µg and purity equivalent or superior to two successive CsCl gradient centrifugations [S2, S3]. The updated Buffer ER formulation excludes Triton X-114, supporting REACH compliance and FDA vaccine standards while reducing the endotoxin-removal incubation from 30 minutes on ice to 5 minutes at room temperature [S2, S3]. The system avoids phenol, chloroform, ethidium bromide, and CsCl [S2, S3].

According to the manufacturer, endotoxin-free preparations show an average transfection efficiency of 154% and 21.0% ± 0.93 percentage of transfected cells, compared with silica-gel slurry methods [S2, S3]. The kit is intended for research on gene therapy and transfection of sensitive cells [S2, S3] and can be stored at room temperature (15–25°C) for up to 2 years [S1].
