Description

Designed for automated cell separation on RoboSep™ instruments, this immunomagnetic negative selection reagent from STEMCELL Technologies depletes red blood cells (RBCs) directly from leukapheresis samples. The kit targets RBCs for removal with antibodies recognizing specific cell surface markers; unwanted cells are labeled with antibodies and magnetic particles, then separated without columns using an EasySep™ magnet. RBC-depleted nucleated cells are simply collected into a new tube and are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Benefits include 99.9% RBC depletion without the need for density gradient centrifugation, sedimentation or lysis, and isolated cells are untouched. With leukapheresis samples, the typical percentage of residual RBCs (Glycophorin A+/CD45-) following use of the reagent is 0.5 ± 0.8 (mean ± SD; n = 15). The reagent is a suspension of magnetic particles and monoclonal antibodies in phosphate-buffered saline (PBS), to be stored at 2 - 8°C and stable until the expiry date (EXP) on the label. To avoid loss of monocytes, EDTA must be added to the leukapheresis sample to a final concentration of 6 mM prior to labeling and separation, and the recommended medium is PBS containing 2% fetal bovine serum, free of Ca++ and Mg++.

For the RoboSep™ protocol, samples of 1 - 5 mL are added to a 14 mL (17 x 95 mm) polystyrene round-bottom tube, the depletion reagent is vortexed for 30 seconds, and the carousel is loaded following the "Human RBC Depletion" on-screen protocol. The product is for research use only and is not suitable for use with downstream magnetic positive selection products.
