EasySep RBC Depletion Reagent - 10mL

Manufactured by STEMCELL Technologies, this immunomagnetic reagent is designed to deplete red blood cells (RBCs) directly from fresh human peripheral blood, umbilical cord blood, bone marrow, buffy coat, or leukapheresis samples. It uses negative selection in which RBCs are targeted with antibodies recognizing specific cell surface markers; unwanted cells are labeled with antibodies and magnetic particles and separated without columns using an EasySep magnet. According to the manufacturer, the reagent achieves 99.9% RBC depletion without the need for density gradient centrifugation, sedimentation, or lysis, and the procedure is fast, easy to use, and column-free, leaving the isolated nucleated cells untouched and immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The product is supplied as a suspension of magnetic particles and monoclonal antibodies in phosphate-buffered saline (PBS) and should be stored at 2 - 8°C (do not freeze). The Safety Data Sheet classifies the mixture as not a hazardous substance or mixture according to the Globally Harmonized System (GHS) and as not considered hazardous by the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200).

Typical residual RBC content (Glycophorin A+/CD45-) reported by the manufacturer is 2 ± 3% for whole blood (mean ± SD; n = 31), 0.39 ± 0.34% for bone marrow (n = 5), 0.12 ± 0.08% for buffy coat (n = 6), 1.7 ± 2.2% for cord blood (n = 16), and 0.5 ± 0.8% for leukapheresis samples (n = 15).
