dPCR CGT Assay WPRE (Cy5)

A wet-lab validated digital PCR assay from QIAGEN targets the woodchuck hepatitis virus post-transcriptional regulatory element (WPRE) sequence for use on the QIAcuity Digital PCR System with the QIAcuity Probe PCR Kit. The Cy5-labeled probe uses a TAO/Iowa Black quencher and yields an 85 bp amplicon. The assay is compatible with the restriction enzymes Msp I, Hpa II, Sma I, and Hae III.

QIAGEN's CGT dPCR assays are validated for both singleplex and multiplex reactions, can be combined with gene-of-interest assays, and are compatible with digital PCR instruments from other suppliers. Accurate quantification is reported down to 0.3 copies/µl, with high linearity demonstrated from 2.5 copies/µl to 15,000 copies/µl and superior precision (<10% deviation from mean) on 8.5k Nanoplates.

The WPRE assay is applied to AAV titer quantification on lysed adeno-associated virus samples, and example data show it being run alongside other CGT dPCR targets such as the CMV promoter and ITR assays in single-well multiplex formats.
