dPCR CGT Assay SV40 polyA (FAM)

QIAGEN's dPCR CGT Assay targets the simian virus 40 polyadenylation signal (SV40 polyA) sequence and is wet-lab validated for digital PCR applications. Designed for use with the QIAcuity Digital PCR System together with the QIAcuity Probe PCR Kit, the assay features FAM fluorophore chemistry with a ZEN/Iowa Black quencher and produces an amplicon length of 95 bp. Sample data demonstrates its application on lysed adeno-associated virus (AAV) samples.

The assay is compatible with the restriction enzymes Msp I, Hpa II, Sma I, and Hae III, and has been validated for both singleplex and multiplex reactions. It can be combined with gene-of-interest assays for AAV genome titer quantification and is also compatible with digital PCR instruments from other suppliers.

Performance characteristics demonstrated on the QIAcuity system include accurate quantification down to 0.3 copies/µl, with grouping of multiple wells enabling detection of rare events. High linearity is shown from 2.5 copies/µl to 15,000 copies/µl, with precision under 10% deviation from the mean on 8.5k Nanoplates.
