dPCR CGT Assay ITR2/5 (Cy5)

A wet-lab validated 20x assay from QIAGEN targets the ITR sequences of adeno-associated virus (AAV) particles. Part of the QIAcuity Cell and Gene Therapy (CGT) dPCR Assay range, it supports viral vector titer measurement, vector genome integrity assessment, and identification of potential plasmid contamination during AAV production, contributing to quality control in gene therapy development workflows.

The assay has an amplicon length of 62 bp and is compatible with the restriction enzymes Msp I, Hpa II, and Sma I. The Cy5 probe uses TAO/Iowa Black as fluorophore and quencher, and a synthetic DNA positive control sequence is supplied. It is designed for use with the QIAcuity digital PCR instrument in combination with the QIAcuity Probe PCR Kit, and is also reported as compatible with digital PCR instruments from other suppliers. The CGT dPCR Assays are validated for singleplex and multiplex reactions and may be combined with gene-of-interest assays.

On 8.5k Nanoplates, the CGT dPCR Assays demonstrate high linearity from 2.5 copies/µl to 15,000 copies/µl with superior precision, reporting less than 10% deviation from the mean.
