dPCR CGT Assay CMV promoter (FAM)

From QIAGEN, this wet-lab validated probe-based assay targets the cytomegalovirus (CMV) promoter sequence and is designed to run on the QIAcuity Digital PCR System using the QIAcuity Probe PCR Kit. A synthetic DNA sequence is included as a positive control. The FAM-labelled probe carries a ZEN/Iowa Black quencher and generates a 128 bp amplicon. The assay is compatible with the restriction enzymes Msp I, Hpa II, Sma I and Hae III, and is validated for AAV vector genome quantification. It can be used in singleplex or multiplex reactions in combination with the CMV enhancer, ITR and SV40 poly A assays, or with gene-of-interest assays, and is also compatible with digital PCR instruments from other suppliers. Demonstrated performance includes high linearity from 2.5 copies/µl to 15,000 copies/µl with precision of <10% deviation from the mean on 8.5k Nanoplates, accurate quantification down to 0.3 copies/µl, and high accuracy across operators and fluorophores.
