dPCR CGT Assay CMV promoter (Cy5)

QIAGEN offers a wet-lab validated, 20x digital PCR assay targeting the cytomegalovirus (CMV) promoter sequence for cell and gene therapy workflows. It is designed for use on the QIAcuity Digital PCR System and is also compatible with digital PCR and quantitative PCR platforms from other suppliers.

The assay has an amplicon length of 128 bp and is compatible with the restriction enzymes Msp I, Hpa II, Sma I, and Hae III. For the Cy5 channel, the probe is configured with a TAO/Iowa Black quencher. A synthetic DNA sequence is provided as a positive control. Multiplex use has been demonstrated on lysed adeno-associated virus (AAV) samples alongside related CGT targets such as SV40 poly A, ITR, and CMV enhancer assays.

QIAGEN's CGT dPCR assay range shows high linearity from 2.5 copies/µl to 15,000 copies/µl with superior precision (less than 10% deviation from mean) on 8.5k Nanoplates, and accurate quantification down to 0.3 copies/µl. The recommended reaction setup uses a volume per reaction of 12 µl within a total reaction volume of 40 µl. The products are intended for molecular biology applications and are not intended for the diagnosis, prevention, or treatment of a disease.
