Description

Designed for genomic DNA isolation from filtered water samples, this QIAGEN kit uses Inhibitor Removal Technology to deliver reliable results in microbial studies, including from turbid water with high levels of contaminants. The workflow removes salts, metals, humic substances and other organic materials that can otherwise interfere with downstream analysis.

Suitable water types include ocean, fresh, brackish, ground, and waste water. After filtration onto a commonly used filter membrane — including 0.45 µm and 0.22 µm filter funnels — the membrane is transferred to a 5 ml PowerWater Bead Pro Tube containing a unique bead mix. Bead-beating lysis occurs by vortexing in a specially formulated lysis buffer. Following protein and inhibitor removal, total genomic DNA is captured on a silica MB Spin Column, washed and eluted in a final 100 µl volume. The purified DNA is ready for downstream applications including PCR, qPCR, Sanger sequencing and next-generation sequencing.

Key specifications include a silica spin filter tube format, bead-beating processing, a binding capacity of up to 20 µg per prep, and a typical prep time of 20 minutes. Kit reagents and components should be stored at room temperature (15–25°C). The product is intended for molecular biology applications. Eluted DNA in Solution EB does not contain EDTA and is recommended for storage at –90°C to –15°C. For higher throughput, a vacuum-based protocol on the QIAvac 24 Plus Manifold supports processing of up to 24 MB Spin Column preps simultaneously, and purification can be fully automated on QIAcube Connect or the classic QIAcube.
