DNeasy Plant Mini Kit (50)

From QIAGEN, this silica-membrane-based kit enables fast and reliable DNA extraction from plant tissues, seeds, and fungi in a spin column format. Designed for purification of total cellular DNA, the resulting nucleic acids are suitable for downstream applications including PCR, qPCR, blotting, and next-generation sequencing. The procedure uses manual processing and can be completed in <1 hour per preparation.

Up to 100 mg of fresh or frozen plant tissue — or 20 mg of dried tissue — can be processed per prep, with a binding capacity of up to 50 µg and an elution volume of 50–200 µl. Typical yields reach up to 30 µg from 50 mg starting material, and purified DNA is sized up to 40 kb. The workflow incorporates a QIAshredder spin column that efficiently removes cell debris and improves sample handling after lysis, while RNA is removed by RNase digestion during lysis. Samples may be fresh, frozen, or dried, and the kit has been used across a broad range of plant species, including wheat, maize, Arabidopsis, tomato, tobacco, rice, oak, and pine. Plant DNA isolation can also be automated on the QIAcube Connect.

Buffer AW1 and Buffer AW2 are supplied as concentrates; ethanol (96–100%) must be added according to the bottle label before use. Lysis and binding buffers contain a chaotropic salt and are not compatible with disinfectants containing bleach.
