DNeasy Plant Maxi Kit (24)

From QIAGEN, this silica-based kit is designed for large-scale plant genomic DNA extraction, with efficient removal of polysaccharides and polyphenols to support downstream PCR and sequencing work [S3]. The workflow is built around a lysis step using Buffer AP1 with RNase A, followed by a Buffer AP2 precipitation step and clarification through a QIAshredder spin column, before DNA binding on a DNeasy Maxi spin column [S1].

Up to 1 g of wet tissue or 0.2 g of lyophilized tissue can be processed per preparation using mechanical disruption with the TissueRuptor, TissueLyser, or a mortar and pestle [S1]. All centrifugation steps are performed at 3000–5000 x g (5000 x g is preferable) at room temperature (15–25°C) in a swing-out rotor [S1]. Elution is carried out in 0.75–1.0 ml Buffer AE [S1]. The kit is suited to demanding applications such as population genetics and whole-genome analysis, delivering robust and reproducible yields [S3].

On arrival the kit should be stored dry at 2–8°C and remains stable for up to 1 year under these conditions [S1].
