Custom LNA Oligonucleotide (1) - 3' TAMRA-Sp

QIAGEN provides custom LNA oligonucleotide synthesis, building Locked Nucleic Acid probes whose bicyclic sugar backbone raises duplex melting temperature relative to standard DNA. Each oligo carries a 3' TAMRA fluorophore attached via a chemical spacer (Sp), eliminating postsynthetic coupling and providing a built-in fluorescence handle for hybridization readouts. TAMRA absorbs maximally near 555 nm and emits around 580 nm, a spectral window that pairs cleanly with common HEX/VIC filter sets on real-time PCR instruments.

Researchers deploy these probes across miRNA in situ hybridization on formalin-fixed sections, dual-probe SNP genotyping assays, mRNA validation by Northern blot, and antisense knock-down screens. Short probe designs of 14 to 20 nucleotides exploit LNA affinity to discriminate single-base mismatches that would escape conventional DNA probes. Each sequence is synthesized on demand at a user-specified yield scale, and every shipment carries mass spectrometry identity confirmation alongside an absorbance-based concentration report.
