Description

From STEMCELL Technologies, this recombinant SARS-CoV-2 spike protein fragment spans amino acids Arg319-Phe541 and is expressed in the yeast Pichia pastoris. The construct carries an N-terminal polyhistidine tag together with a SUMOstar site and corresponds to a region of the spike (S) glycoprotein, one of the four structural proteins encoded by the SARS-CoV-2 genome. The S1 subunit contains the receptor-binding domain that engages the human ACE2 receptor, and this fragment is used to study viral attachment and to support monoclonal antibody and vaccine development.

The protein is supplied lyophilized from a solution containing Tris-HCl, NaCl, and trehalose at pH 8.0. Its predicted molecular mass is 38.1 kDa. The product is glycosylated; on SDS-PAGE the band migrates at approximately 68 kDa before Endoglycosidase H treatment and at approximately 38 kDa after deglycosylation. Purity assessed by SDS-PAGE is reported as ≥ 90%.

Binding activity was determined by functional ELISA using immobilized SARS-CoV-2 Recombinant Spike Protein together with Human Recombinant ACE2 Protein, yielding an EC50 of ≤ 37 ng/mL. The lyophilized material should be stored at -20°C to -80°C, reconstituted in sterile water to a concentration of 0.1 - 1.0 mg/mL by pipetting (without vortexing), sterile-filtered at 0.2 μm, and held at 2 - 8°C for up to one week.
