Description

QIAGEN supplies this set of controls for use alongside Cignal Reporter Assays, where proper controls are a key element of the experimental design. They permit accurate interpretation of reporter assay results and provide assurance of the specificity of the observed response. The reagents are supplied as preformulated, transfection-ready constructs.

Positive and negative controls are available for both dual-luciferase and GFP reporters. The Positive Control Assay (luc) is a 40:1:1 mixture of constitutively expressing GFP, constitutively expressing firefly luciferase, and constitutively expressing Renilla luciferase constructs, allowing measurement of transduction efficiency and serving as a positive control for firefly luciferase assays. The Negative Control Assay (luc) is a 40:1 mixture of a non-inducible firefly luciferase construct and a constitutively expressing Renilla luciferase construct, used to establish the specificity of treatment effects. For GFP-based work, the Positive Control Assay (GFP) constitutively expresses GFP, making it suitable for measuring transduction efficiency and optimising transduction conditions, while the Negative Control Assay (GFP) uses a noninducible, minimal promoter-driven GFP gene to establish specificity and determine background GFP fluorescence.

Cignal Lenti Reporter Control Assays are also available as pretitered, transduction-ready lentivirus particles for optimising transduction conditions, with control-transduced cells treated in the same way as reporter-transduced cells.
