CGT Viral Vector Lysis kit (1000 rxn)

This direct lysis kit from QIAGEN is designed for use in cell and gene therapy workflows, providing standardized sample processing for the lysis of adeno-associated virus (AAV) vectors and adenoviruses. The kit is suitable for the lysis of AAV2, AAV5, AAV6, AAV8, and AAV9 serotypes, with lysates optimized for use with QIAcuity Cell and Gene Therapy dPCR Assays in combination with the QIAcuity Probe PCR Kit.

The kit integrates with the QIAcuity Digital PCR System and QIAcuity Nanoplate 8.5K to deliver an end-to-end dPCR workflow providing absolute quantification of vector genome copies, with consistent, robust, and accurate determination of viral titer while saving hands-on time compared with workflows requiring a separate sample preparation step. Restriction enzymes such as Hpall referenced in the protocol are not included.

In validation, two AAV2 samples were processed with this kit and quantified on the QIAcuity Digital PCR instrument using 8.5k Nanoplates and CGT dPCR Assays run in triplex reactions in the FAM, HEX, and Cy5 channels. Samples were serially diluted in six steps from 15,000 copies/µL down to 2.5 copies/µL (R2=1.0), with each dilution measured in technical triplicates and quantifications performed both with (+) and without (–) restriction digest of the ITR regions.
