Description

A plant-derived polyphenol and tetrahydroxychalcone supplied by STEMCELL Technologies, this small molecule acts as an NF-κB pathway inhibitor that stabilizes IκB. The compound prevents phosphorylation and degradation of TNF-dependent IκBα, an inhibitory subunit of NF-κB (IC₅₀ = 38 µM). It also inhibits 5-lipoxygenase (IC₅₀ = 0.01 µM), the enoyl-acyl-carrier protein reductase of Plasmodium falciparum (Ki = 2.97 µM), angiotensin-converting enzyme (IC₅₀ = 0.73 mM), and SRC kinase.

Product details include a chemical name of (2E)-1-(2,4-dihydroxyphenyl)-3-(3,4-dihydroxyphenyl)-2-propen-1-one, molecular formula C₁₅H₁₂O₅, molecular weight of 272.3 g/mol, and a purity of ≥ 95%. It is supplied as a crystalline solid. Recommended storage is at -20°C, protected from prolonged exposure to light; the product is stable as supplied for 12 months from date of receipt. Solubility is reported at ≤ 70 mM in DMSO and ≤ 70 mM in ethanol. The compound has low solubility in aqueous media; when used as a cell culture supplement, the stock solution should be diluted into culture medium immediately before use, with final DMSO concentration kept below 0.1% to avoid potential cell toxicity.

Published research applications span metabolism (inhibition of iron-induced lipid peroxidation in rat brain homogenate), cancer research (inhibition of TNF-α-induced invasion in human lung adenocarcinoma H1299 cells, growth inhibition in HepG2 and Hep3B hepatoma cell lines via G2/M phase arrest, and inhibition of testosterone-induced proliferation in breast cancer cells), and immunology (anti-inflammatory effects in a mouse macrophage cell line through inhibition of LPS-induced expression of inducible nitric oxide synthase).
