Description

A plant polyphenol and tetrahydroxychalcone from STEMCELL Technologies, Butein is characterized as an NF-κB pathway inhibitor that stabilizes IκB. It inhibits nuclear factor (NF)κB and has been shown to prevent phosphorylation and degradation of TNF-dependent IκBα, an inhibitory subunit of NFκB, with an IC₅₀ of 38 µM. The compound also inhibits 5-lipoxygenase (IC₅₀ = 0.01 µM), the enoyl-acyl-carrier protein reductase of Plasmodium falciparum (Ki = 2.97 µM), angiotensin-converting enzyme (IC₅₀ = 0.73 mM), and SRC kinase.

Supplied as a crystalline solid with a purity of ≥ 95%, the compound has a molecular formula of C₁₅H₁₂O₅ and a molecular weight of 272.3 g/mol. Its chemical structure corresponds to 2',3,4,4'-tetrahydroxychalcone. It is soluble in DMSO and ethanol at concentrations up to 70 mM, and is stable at -20°C as supplied. For long-term storage, it should be kept with a desiccant and protected from prolonged exposure to light; one source indicates stability for 12 months from the date of receipt.

Published research applications include metabolism, where the compound inhibits iron-induced lipid peroxidation in rat brain homogenate in a concentration-dependent manner; cancer research, where it inhibits TNF-α-induced invasion of human lung adenocarcinoma H1299 cells, growth in human hepatoma cancer cell lines HepG2 and Hep3B by inducing G2/M phase arrest, and testosterone-induced proliferation in breast cancer cells; and immunology, where it exhibits anti-inflammatory properties in a mouse macrophage cell line by inhibiting LPS-induced expression of iNOS. Products from STEMCELL Technologies are for research use only and not intended for human or animal diagnostic or therapeutic uses.
