Anti-ZO1 Antibody

The recombinant rabbit monoclonal reagent distributed by Antibodies.com targets ZO1 (Zonula Occludens-1), a ~220 kDa MAGUK-family scaffolding protein that organizes the cytoplasmic plaque of tight junctions in epithelial and endothelial tissues.

Supplied as 100 µL of unconjugated antibody in TBS (pH 7.4) with 1% BSA, 40% glycerol, and 0.05% sodium azide as preservative, the clone is validated for immunocytochemistry (ICC) and immunohistochemistry (IHC) on human, mouse, and rat samples. It binds the cytoplasmic face of the apical junctional complex, where ZO1 anchors transmembrane partners such as claudins, occludin, and JAM proteins to the actin cytoskeleton through its PDZ, SH3, and guanylate kinase domains.

Researchers routinely apply this antibody to stain the tight junction belt of polarized monolayers such as Caco-2, MDCK, and primary enterocyte cultures, to chart blood-brain barrier maturation in rodent brain sections, and to score epithelial-mesenchymal transition in tumor and tissue injury models. Because ZO1 redistributes from the membrane to the cytosol when paracellular barrier function is compromised, it serves as a sensitive readout for permeability changes induced by cytokines, pathogens, or experimental knockdown of junction components. In IHC workflows the unconjugated format lets the user pair the primary with any species-matched secondary antibody and detection chemistry (HRP, fluorescence, or chromogenic) appropriate to the experiment.

For storage, aliquot upon receipt and store at -20°C in a manual defrost freezer to preserve activity through repeated freeze-thaw cycles; the 40% glycerol formulation also tolerates short-term refrigerated storage. Each vial ships at 100 µL, a working volume sufficient for multiple immunostaining runs depending on dilution and section thickness.
