Anti-ZAP70 Antibody

Offered by Antibodies.com within its Primary Antibodies portfolio, this rabbit polyclonal reagent targets zeta-chain-associated protein kinase 70 (ZAP70), a 70 kDa Syk-family tyrosine kinase that couples the T-cell receptor to downstream signaling cascades. Supplied as 100 µl of unconjugated antiserum, it is validated for western blot (WB), immunohistochemistry (IHC), immunofluorescence (IF), and immunoprecipitation (IP) workflows using Human, Mouse, and Rat samples.

ZAP70 is recruited to doubly phosphorylated ITAM motifs within the CD3 complex, where it phosphorylates the adaptor LAT and contributes to downstream calcium flux and MAPK pathway activation. Loss-of-function mutations in the encoding gene cause autosomal recessive severe combined immunodeficiency (SCID), while aberrant ZAP70 expression has been documented in B-cell chronic lymphocytic leukemia (B-CLL) and in selected autoimmune contexts, supporting its relevance to immunology, hematology, and signal-transduction research.

The polyclonal host background enables recognition of multiple epitopes on the ZAP70 protein, which is useful when probing phosphorylation-dependent conformational states. The unconjugated format allows end users to couple the antibody to HRP, biotin, fluorophores, or beads of their choosing. For handling, the vial should be stored at -20 °C and aliquoted to limit repeated freeze-thaw cycles of the immunoglobulin preparation.
