Produced in rabbit, this unconjugated polyclonal reagent from Antibodies.com recognizes XRCC4, a scaffolding protein of the non-homologous end joining pathway that reseals DNA double-strand breaks in coordination with Ku70, Ku80, and DNA Ligase IV. The antibody is supplied as a 100 µl volume and has been validated for Western blot and immunohistochemistry on human, mouse, and rat samples.

XRCC4 is essential for V(D)J recombination during lymphocyte receptor assembly, and loss-of-function mutations in the encoding gene are linked to radiosensitive severe combined immunodeficiency phenotypes. Researchers apply this reagent to study DNA damage responses, tumor cell radiosensitivity, genomic instability in cancer models, and the mechanisms of class-switch recombination in B cells, including work on microhomology-mediated end joining as a backup repair route.

The unconjugated format leaves investigators free to couple their own reporter tags, including HRP, biotin, or fluorophores, and the polyclonal host response offers recognition of multiple epitopes across the XRCC4 sequence, which can be helpful when probing truncated fragments generated during apoptosis, chromatin fractionation, or in vitro nuclease cleavage assays.
