Produced in rabbits and affinity-purified from immune serum, this unconjugated polyclonal IgG reagent binds the TSTA3 protein (also annotated as FX or GDP-L-fucose synthase), the NADP-dependent epimerase-reductase that completes the de novo biosynthetic pathway converting GDP-mannose to GDP-L-fucose. Functional TSTA3 activity supplies the nucleotide-sugar donor required by cellular fucosyltransferases to assemble selectin ligands and to modify secreted or surface glycoproteins; biallelic loss-of-function alleles in this gene cause leukocyte adhesion deficiency type II, a congenital disorder of glycosylation that features the Bombay erythrocyte phenotype, chronic neutropenia, and recurrent infections. Researchers therefore apply this reagent to monitor induction or suppression of fucose pathway flux in tumor cell migration models, to compare basal and stimulated glycosylation during immune cell maturation, to validate CRISPR knockouts and matched rescue controls, and to characterize patient-derived tissue lysates.

Validated by Antibodies.com for Western blot, the antibody recognizes endogenous total TSTA3 and produces a band near 36 kDa on SDS-PAGE from human and mouse lysates. The 100 µl liquid aliquot accepts HRP, biotin, or fluorescent secondary conjugates for chemiluminescent, chromogenic, or near-infrared readout on PVDF or nitrocellulose membranes. Because the polyclonal format targets multiple epitopes of the approximately 321-amino-acid TSTA3 polypeptide, the reagent retains reactivity against partially degraded targets and against low-abundance samples typical of glycoprotein biochemistry workflows. Intended for research use only and not validated for diagnostic applications.
