Raised in rabbit, this unconjugated polyclonal antibody recognizes TRIM63, the E3 ubiquitin-protein ligase commonly referred to as MuRF1. MuRF1 is a central regulator of skeletal and cardiac muscle protein turnover, targeting myofibrillar substrates such as myosin heavy chain, troponin I, and myosin-binding protein C for ubiquitin-dependent degradation during atrophy. The reagent from Antibodies.com is supplied at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, packaged as a 100 µl vial, and has been validated for western blot, immunohistochemistry, and immunofluorescence workflows on human, mouse, and rat specimens.
TRIM63 expression rises sharply in skeletal muscle under catabolic conditions including denervation, immobilization, glucocorticoid treatment, fasting, cancer cachexia, sepsis, and chronic heart failure, and the same ligase has been linked to cardiac hypertrophy and proteasomal remodeling in cardiomyocytes. Researchers use Anti-TRIM63 antibodies to mark atrophying myofibers, to quantify MuRF1 induction after disuse or exercise interventions, to probe sarcomere assembly during myogenesis, and to assess cardiac stress responses in rodent disease models. The polyclonal format provides broader epitope coverage that can be useful when probing both full-length TRIM63 and the shorter isoforms reported in differentiating myotubes.
For western blot, run reduced samples and detect the expected band near 40 kDa, with occasional higher-molecular-weight species corresponding to dimeric or post-translationally modified MuRF1. For immunofluorescence and immunohistochemistry on frozen or formalin-fixed sections, antigen retrieval with citrate buffer at pH 6 followed by an indirect detection protocol typically yields clear cytoplasmic and sarcomeric staining in striated muscle. Storage at -20 °C in working aliquots is recommended to limit freeze-thaw cycles of the glycerol-containing stock, while short-term preservation at 4 °C is acceptable thanks to the included sodium azide. This reagent is intended for research use only and is not validated for diagnostic or therapeutic applications.
