For lysosomal storage disorder and neurodegeneration research, this rabbit polyclonal IgG reagent from Antibodies.com targets human tripeptidyl peptidase 1 (TPP1), a lysosomal serine protease best known for its central role in CLN2 disease (neuronal ceroid lipofuscinosis type 2, also called classic late-infantile Batten disease). TPP1 normally cleaves N-terminal tripeptides from polypeptides delivered to the lysosome and is widely studied in enzyme replacement therapy work and TPP1-deficient model systems.

Supplied as 100 µL of unconjugated antiserum in a single-use vial, the antibody is validated for Western blot detection of endogenous human TPP1. Its polyclonal format recognizes multiple epitopes on the TPP1 protein, which is useful when probing lysosomal fractions, fibroblast or neuronal cell lysates, and brain tissue preparations where TPP1 expression and processing can be monitored.

Typical workflows include lysate preparation in RIPA or non-ionic detergents, SDS-PAGE separation, transfer to nitrocellulose or PVDF, and chemiluminescent or fluorescent readout. Because TPP1 runs as a pro-form (~68 kDa) that is processed in the lysosome to a mature heavy chain of ~46 kDa plus a light chain of ~20 kDa, Western blot with this reagent can resolve both precursor and mature species in patient fibroblasts, iPSC-derived neurons, and TPP1-deficient models, supporting mutation studies and genotype-phenotype work.
