This rabbit polyclonal antibody from Antibodies.com targets TOB1 (Transducer of ERBB2, 1), a member of the BTG/Tob family of antiproliferative proteins that regulate G1/S cell cycle progression and modulate Smad-mediated transcription downstream of BMP and TGF-beta signaling. The unconjugated IgG reagent is supplied at a 200 ul pack size and has been validated for Western blot, immunohistochemistry, and flow cytometry on human and mouse samples.

TOB1 functions as a tumor suppressor in several biological contexts: it dampens ERBB2-driven MAPK signaling, recruits poly(A)-binding protein to control cytoplasmic mRNA deadenylation, and limits proliferation of osteoblasts and resting T lymphocytes. Investigators working on bone formation, immune cell quiescence, or ErbB2-related oncogenic pathways use this reagent to confirm knockdown efficiency, follow endogenous expression, or localize the protein in fixed tissue sections.

Storage and handling: aliquot the antiserum upon arrival to limit freeze-thaw cycles and store at -20 degrees C; protect from light. For Western blot, dilute in 5% non-fat milk or BSA in TBST. For IHC on formalin-fixed paraffin-embedded tissue, perform heat-induced antigen retrieval in citrate buffer at pH 6.0. For flow cytometry, titrate on the relevant cell line. The polyclonal pool recognizes epitopes distributed across full-length TOB1 and is intended for research use only.
