The Toa1 protein in Saccharomyces cerevisiae is the large subunit of the general transcription factor TFIIA, pairing with the small subunit Toa2 to stabilize TATA-binding protein (TBP) on core promoters and assist RNA polymerase II during transcription initiation. This rabbit IgG polyclonal reagent, distributed by Antibodies.com, is supplied as a liquid at 100 µl and is validated for western blot detection of endogenous Toa1 in yeast whole-cell extracts.

Research applications

  • Studies of TFIIA assembly, TBP-promoter recruitment, and pre-initiation complex formation
  • Characterization of transcription factor mutants and suppressor screens in S. cerevisiae
  • Western blot analysis of yeast lysates prepared by TCA precipitation or glass-bead mechanical disruption

Handling and storage

Store at -20 °C in single-use aliquots, protected from light, and limit freeze-thaw cycles to preserve binding activity. For western blot, dilute the antibody 1:500 to 1:2000 in blocking buffer; the resulting band appears near the predicted ~27 kDa Toa1 polypeptide on standard SDS-PAGE gels. Intended for research use only in non-clinical laboratory workflows.
