The Antibodies.com polyclonal reagent specifically recognizes Transition Protein 2 (TNP2), a small, highly basic nuclear protein expressed in haploid round and elongating spermatids of the testis. TNP2 works together with TNP1 to displace canonical histones and facilitate the transient chromatin remodeling that precedes final protamine-driven condensation in mature spermatozoa, so antibodies against TNP2 are widely used to study spermatid differentiation, male germ cell maturation, and chromatin repackaging events relevant to male fertility research.

This affinity-purified polyclonal antibody was raised in rabbit and is supplied as an unconjugated preparation at 1 mg/ml in phosphate buffered saline (PBS), pH approximately 7.2, with 15 mM sodium azide as a preservative. The shipping volume is 100 µl, enough for multiple western blot experiments on testis or germ cell extracts. Antibody reactivity has been verified against human and rat samples, and the recommended application is Western blot detection of endogenous TNP2 in denaturing SDS-PAGE lysates prepared from testicular tissue or isolated germ cell fractions. For best stability, keep the vial refrigerated after arrival, protect it from light, and avoid repeated freeze-thaw cycles by working from small aliquots.

Typical experimental use includes probing nitrocellulose or PVDF membranes carrying separated testis proteins with this reagent, then developing the blot with an appropriate HRP- or fluorescent-labeled secondary antibody to visualize stage-specific TNP2 bands. Because TNP2 appears only transiently during a narrow window of spermiogenesis, the antibody is also useful for tracking the progression of spermatid maturation in knock-out, knock-down, or chemically treated animal models of male infertility.
