TNFRSF12A (also known as FN14, TWEAKR, or CD266) is a type I transmembrane protein of the tumor necrosis factor receptor superfamily that serves as the sole signaling receptor for the ligand TWEAK (TNFSF12). Engagement of TNFRSF12A activates NF-κB, MAPK, and caspase-dependent pathways, driving context-dependent outcomes in proliferation, migration, angiogenesis, and apoptosis. Its expression is low in healthy adult tissue but rises sharply during tissue injury, chronic inflammation, and in a broad spectrum of solid tumours, making reliable immunological detection a priority across oncology and regenerative-medicine research programmes.
This polyclonal reagent from Antibodies.com is raised in rabbit and supplied as an unconjugated, 100 µL liquid aliquot in a single-use vial, suitable for direct labelling or secondary detection workflows. The antibody has been validated on human samples for Western blot analysis of cell and tissue lysates, where it recognises the receptor under reducing conditions, and for immunohistochemistry on formalin-fixed, paraffin-embedded sections, where it reveals membranous and cytoplasmic staining patterns consistent with the known subcellular distribution of TNFRSF12A.
Typical laboratory uses include profiling receptor upregulation in glioblastoma, breast, prostate, hepatocellular, and pancreatic carcinoma models, characterising FN14/TWEAK signalling in wound-healing and fibrotic disease studies, and tracking TNFRSF12A induction in inflammatory contexts such as rheumatoid synovium, atherosclerotic plaques, and ischemia-reperfusion injury. Researchers preparing whole-cell lysates in RIPA buffer, resolving proteins by SDS-PAGE, and transferring to PVDF or nitrocellulose membranes will find the antibody compatible with standard chemiluminescent detection, while IHC users can employ antigen retrieval in citrate or EDTA buffer followed by HRP- or AP-conjugated anti-rabbit secondaries. The unconjugated format gives laboratories flexibility to pair the reagent with their preferred fluorescent, biotinylated, or enzymatic secondary antibodies.
