Raised in rabbit against the transmembrane protein with EGF-like and two follistatin-like domains 2 (TMEFF2), this unconjugated polyclonal IgG reagent from Antibodies.com is supplied as a 50 µl vial and validated for western blot detection of endogenous protein in human, mouse, and rat samples. TMEFF2 is a 374-residue type-I transmembrane proteoglycan encoded on chromosome 2q32.3 that carries an N-terminal signal peptide, two follistatin-like repeats, an EGF-like module, a transmembrane region, and a short cytoplasmic tail bearing a putative G-protein activation motif. The predicted, unglycosylated molecular mass is approximately 41 kDa, although the heavily glycosylated and proteolytically processed forms migrate at varied apparent weights on SDS-PAGE. The follistatin-like domains sequester TGF-beta superfamily ligands and modulate BMP signalling, while the ectodomain is shed from the cell surface through ADAM17 and gamma-secretase activity. Investigators apply this reagent in studies of prostate cancer, where TMEFF2 acts as a candidate tumor suppressor that is frequently silenced by 5-prime CpG-island hypermethylation, as well as in work on hippocampal and mesencephalic neuron survival, amyloid-beta binding relevant to Alzheimer's disease, and integrin/RhoA-mediated cell migration. Lysates are commonly prepared in RIPA or NP-40 buffer under reducing conditions, separated by SDS-PAGE, and transferred to membranes for probing, with the unconjugated IgG pairing flexibly with HRP- or fluorophore-labelled secondary reagents. Store at -20 °C as a frozen aliquot and limit freeze-thaw cycles to preserve polyclonal reactivity. For research use only.
