Rerotic, calcium-dependent transglutaminase 1 (TGM1) is a keratinocyte-enzyme that catalyzes isopeptide cross-links between glutamine and lysine residues of structural proteins during the final stages of epidermal differentiation, building the cornified cell envelope that gives the stratum corneum its mechanical and barrier integrity. Loss-of-function variants in the TGM1 gene cause autosomal recessive lamellar ichthyosis, and altered TGM1 expression is also reported in psoriasis, atopic dermatitis, and several epithelial tumor contexts, making the protein a long-standing target for dermatology and skin biology research.
