Anti-Tau (phospho Thr181) Antibody [2A4]

Offered through Antibodies.com, this mouse monoclonal reagent (clone 2A4) targets the Tau protein phosphorylated at threonine 181 and is supplied at 500 µg per vial in a lyophilised format, prepared from a 0.01 M PBS (pH 7.2) solution and intended to be reconstituted in double-distilled water to a working concentration of about 1.00 mg/mL. The antibody is of the IgG2a, κ isotype, is produced in the Sp2/0-Ag14 myeloma line, and is unconjugated, allowing it to be paired with the secondary detection reagent of choice. It was raised against a KLH-conjugated phosphopeptide spanning residues 177–187 of human Tau (APKTpPPSSGGC), Protein G affinity purified, and validated for Western blot, where it consistently detects an approximately 46 kDa band in mouse brain lysate at a working dilution of 1:2000 (≈0.5 µg/mL). Reactivity has been confirmed on Human and Mouse samples, in keeping with the broader species profile of the target, and cross-reactivity with the un-phosphorylated version of the same peptide should be taken into account during assay design. For long-term integrity the lyophilised product should be stored at −20 °C, with repeated freeze–thaw cycles avoided after reconstitution.
