Phosphorylation of the microtubule-associated protein tau at serine 396 is a late-stage pathological modification deposited in paired helical filaments and neurofibrillary tangles characteristic of Alzheimer's disease and related tauopathies. This unconjugated rabbit polyclonal IgG reagent from Antibodies.com was raised against a phospho-Ser396 peptide and affinity-purified by antigen chromatography, so it selectively recognizes endogenous tau only when residue 396 is phosphorylated and does not bind the unmodified protein. It is validated for western blot and immunohistochemistry on samples of human, mouse, and rat origin, giving cross-species coverage for transgenic rodent models and human post-mortem brain tissue studies.

The reagent is supplied as 100 µL of liquid in a single-use vial. For optimal phospho-epitope integrity, aliquot the stock on arrival, store working aliquots frozen at -20 °C, and limit freeze-thaw cycles. Researchers commonly pair this reagent with anti-total tau and other site-specific phospho-tau antibodies to map kinase-driven tau hyperphosphorylation in cell and animal models, stage tangle pathology in fixed brain sections, and probe how site-specific phosphorylation correlates with seeding-competent tau aggregates. For research use only.
