Phospho-specific detection of Synapsin-1 at serine 549 is supported by this rabbit polyclonal IgG reagent from Antibodies.com, supplied as 200 µl of unconjugated antibody validated for Western blot and reactive against human, mouse, and rat samples. The antibody discriminates the PKA/CaMKII-phosphorylated form of Synapsin-1 from the non-phosphorylated protein, enabling studies of activity-dependent synaptic vesicle mobilization in primary neuron cultures and rodent brain tissue.

Synapsin-1 is a presynaptic phosphoprotein that tethers synaptic vesicles to the actin cytoskeleton. Phosphorylation at S549 lowers its affinity for vesicles and shifts them into the actively recycling pool during sustained firing. Probing membrane transfers with this reagent therefore reports on the dynamic, mobilized fraction of Synapsin-1, complementing total-Synapsin-1 blots that reflect only steady-state protein levels and offering a biochemical readout of presynaptic activity over time.

For Western blot, separate samples by SDS-PAGE, transfer to nitrocellulose or PVDF, block, and incubate with the antibody before detecting bound immunoglobulin with an HRP-conjugated anti-rabbit IgG secondary and chemiluminescent substrate. Aliquot upon arrival to avoid repeated freeze-thaw cycles, store refrigerated at 4°C in the supplied vial, and keep on ice during handling to preserve phospho-specific binding. The reagent is supplied in a single-use liquid format intended for research use only and is not validated for diagnostic applications.
