The Anti-Syk (phospho-Y323) Antibody from Antibodies.com is a rabbit polyclonal reagent that selectively recognizes spleen tyrosine kinase (Syk) when phosphorylated at the tyrosine 323 autophosphorylation site. Syk is a 72 kDa non-receptor cytoplasmic tyrosine kinase that operates downstream of B-cell receptors, Fc receptors, and integrin-based adhesion complexes, linking immune receptor engagement to calcium mobilization, cytoskeletal reorganization, and transcriptional activation in B lymphocytes, mast cells, macrophages, neutrophils, and platelets.

Validated for western blot and immunohistochemistry on human, mouse, and rat samples, the antibody is supplied at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide at approximately pH 7.2, packaged in a 100 µl vial. The unconjugated format is compatible with a broad selection of secondary detection reagents and labeling chemistries, supporting phospho-proteomics, signaling pathway studies, and immune cell research workflows where receptor-proximal kinase activation needs to be resolved from total protein levels.

Tyrosine 323 has been characterized in the literature as a regulatory autophosphorylation event that modulates Syk catalytic output and downstream substrate recruitment, making this reagent useful for tracking receptor-proximal kinase activation across hematopoietic lineages and innate immunity contexts. For short-term use, store refrigerated and protected from light; for longer-term storage, aliquot and freeze, avoiding repeated freeze-thaw cycles to preserve binding activity.
