Raised in rabbits against Small Ubiquitin-like Modifier 4, this unconjugated polyclonal IgG reagent from Antibodies.com is supplied as 200 µl of liquid in a single-use vial and validated for western blot and immunofluorescence workflows on human, mouse, and rat samples. SUMO4 is an approximately 18 kDa ubiquitin-like modifier that covalently attaches to lysine residues on substrates such as IκBα, dampening NF-κB-dependent transcription and tying sumoylation to immune signalling. The reagent pairs with HRP- or fluorophore-conjugated secondary antibodies for chemiluminescent western blot band detection at the mature 18 kDa position and for clean nuclear or cytoplasmic staining on cultured cells and tissue cryosections. Researchers apply it to probe the M55V (A163G) polymorphism, which has been repeatedly associated with type 1 diabetes susceptibility in Asian and European cohorts, and to map stress-dependent conjugation of antioxidant enzymes, chaperones, and stress defence proteins under oxidative challenge. Because Pro-90 renders SUMO4 insensitive to sentrin-specific proteases, the unconjugated reagent is also useful in side-by-side comparisons with SUMO1/2/3 conjugates. For stability, the vial should be stored at -20 °C in working aliquots and protected from light to preserve activity through limited freeze-thaw cycles, and each lot is affinity purified and quality controlled for specificity toward the SUMO4 target.
