Researchers probing post-translational SUMO conjugation use this rabbit polyclonal IgG reagent from Antibodies.com to detect Small Ubiquitin-like Modifier 4 in human, mouse, and rat samples. SUMO4 is a 95 amino acid modifier that shares roughly 87% sequence identity with SUMO2, and contemporary validation studies caution that anti-SUMO4 polyclonal antibodies often cross react with SUMO2 and SUMO3 conjugates due to the conserved C-terminal region, so pairing with SUMO2/3 knock-down or orthogonal RNAi controls is advisable for clean interpretation of high molecular weight bands.
The unconjugated antibody is validated for western blot and immunofluorescence. Recommended starting dilutions range from 1:1000 to 1:2000 for western blot and from 1:100 to 1:300 for immunofluorescence on cultured cells and tissue sections, allowing the same vial to support multiple nitrocellulose membranes and coverslip staining sessions. Investigators employ it to chart SUMO4 expression in immune tissues such as lymph node and spleen, in adult and embryonic kidney, and in pancreatic islets where the SUMO4 M55V polymorphism has been linked to type 1 diabetes susceptibility through altered conjugation of IκBα and dampening of NF-κB dependent IL12B transcription. The reagent is also useful for mapping how SUMO4 modifies antioxidant enzymes, chaperones, and stress defense proteins under oxidative challenge in renal and hepatic cell lines.
Supplied as a 100 µL liquid in a vial containing phosphate buffered saline with glycerol and stabilizing proteins, the antibody should be aliquoted on first thaw and stored at -20°C to limit freeze-thaw cycles of the IgG stock. For chemiluminescent detection, pair the primary with an HRP conjugated goat anti-rabbit IgG secondary; for multiplex immunofluorescence, pair with species-matched Alexa Fluor or DyLight conjugates.
