The 4D12 clone is a rat monoclonal antibody that specifically recognizes SUMO1 (Small Ubiquitin-like Modifier 1), an ~11.5 kDa ubiquitin-like protein that is covalently conjugated to lysine residues on target substrates to regulate nuclear transport, transcription, apoptosis, and protein stability. The biotinylated formulation from Antibodies.com enables amplification of signal when paired with streptavidin-HRP or streptavidin-fluorophore conjugates.

This reagent is supplied as a liquid at 50 µg per vial and is validated across multiple assay platforms including Western blot, immunofluorescence, frozen-section immunohistochemistry, and ELISA on human, monkey, mouse, and rat samples. For Western blot, a starting dilution of 1:500 to 1:2000 is commonly recommended; titrate empirically for immunofluorescence staining on fixed cells or tissue cryosections.

The 4D12 epitope reportedly detects both free unconjugated SUMO1 and SUMO1-conjugated substrates, making it useful for studies of the sumoylation machinery in cell lysates and tissue sections. Aliquot upon receipt to avoid repeated freeze-thaw cycles; refrigerate at 4°C for short-term use or store frozen at -20°C for longer-term stability.
