The SUFU protein (Suppressor of Fused homolog) acts as a critical negative regulator of the Hedgehog (Hh) signaling pathway, binding GLI transcription factors and retaining them in the cytoplasm to prevent their nuclear accumulation and downstream gene activation. Loss-of-function mutations and altered expression of SUFU are associated with developmental patterning defects and with malignancies such as medulloblastoma and basal cell carcinoma, which has positioned the protein as a frequent subject in cancer biology and signal transduction studies.

This unconjugated rabbit polyclonal antibody from Antibodies.com was raised against human SUFU and is validated for Western blot (WB) and immunohistochemistry (IHC). It detects the endogenous human protein and is supplied as 100 µl of polyclonal antiserum, ready for use on cell lysates and fixed tissue sections. Investigators examining Hedgehog pathway activity, GLI factor dynamics, or tumor suppressor behavior in human samples can pair this reagent with genetic, transcriptomic, or pharmacologic readouts to confirm protein-level changes.

For Western blot, samples are typically resolved on SDS-PAGE gels and transferred to nitrocellulose or PVDF membranes, then probed with the antibody at empirically determined dilutions. For IHC, the reagent is compatible with standard formalin-fixed, paraffin-embedded (FFPE) protocols following appropriate antigen retrieval and horseradish peroxidase- or fluorophore-based detection. Storage at -20 °C in single-use aliquots helps preserve antibody performance and limit freeze-thaw cycles across repeated experiments.
