STX4 (Syntaxin-4) is a plasma-membrane t-SNARE that anchors the docking and fusion of secretory vesicles with the cell surface, and the unconjugated rabbit polyclonal IgG reagent from Antibodies.com is raised against this integral membrane SNARE. Supplied as a 50 µl liquid vial, the antibody is validated for western blot detection of endogenous STX4 in human, mouse, rat, and monkey samples, and the polyclonal format increases the chance of recognizing multiple epitopes on denatured target in transfected cell lysates or primary tissue homogenates.

STX4 assembles into a four-helix SNARE bundle with the partner proteins SNAP-23 or SNAP-25 on the plasma membrane and vesicular VAMP2 or related VAMP isoforms, and the complex drives membrane fusion events central to insulin-stimulated GLUT4 translocation in adipocytes and skeletal muscle, dense granule release in platelets, and calcium-triggered exocytosis at neuronal and neuroendocrine synapses. Research applications of this reagent include characterizing SNARE complex assembly and insulin signaling in metabolic disease models, mapping the contribution of STX4 to platelet degranulation in thrombosis and hemostasis studies, and probing the role of presynaptic syntaxin isoforms in neurotransmitter release.

For western blot, the antibody is commonly used at working concentrations in the 0.2-1 µg/ml range with HRP-conjugated anti-rabbit secondary antibodies, and the unconjugated format allows investigators to pair the primary with their preferred reporter system. Store at -20 °C in working aliquots to limit repeated freeze-thaw cycles of the IgG preparation, and gently mix before each use. Optimal working dilutions should be determined empirically for each lysate type and detection chemistry.
