Anti-STAT5a (Ab-694) Antibody

This rabbit polyclonal reagent from Antibodies.com specifically recognizes STAT5a when phosphorylated at tyrosine 694 (Tyr694), a key activating site within the C-terminal transactivation domain. Supplied unconjugated in a 100 µl volume, the antibody is validated for Western blot (WB) and immunohistochemistry (IHC) workflows on human, mouse, and rat samples.

STAT5a is a latent cytoplasmic transcription factor of the JAK-STAT signaling pathway. Cytokine- and growth-factor-driven phosphorylation at Tyr694 triggers dimerization, nuclear translocation, and induction of gene programs involved in proliferation, differentiation, and immune cell development. Detection of phospho-Tyr694 is therefore a direct readout of pathway activation in response to stimuli such as IL-2, IL-3, IL-5, IL-7, IL-15, GM-CSF, erythropoietin, thrombopoietin, and prolactin, supporting studies of hematopoietic lineage commitment, T-cell and NK-cell biology, mammary gland development, and certain leukemia models.

For Western blot, the reagent can probe whole-cell lysates prepared with standard RIPA buffer plus phosphatase inhibitors to preserve phosphorylation. For immunohistochemistry, it supports localization studies on formalin-fixed paraffin-embedded or frozen tissue sections.
