SQSTM1, also called p62 or sequestosome-1, is a multidomain scaffolding protein that functions as a selective autophagy receptor. It binds ubiquitinated cargo through its C-terminal UBA domain and tethers that cargo to LC3-positive autophagosomes via its LIR motif. Because SQSTM1 itself is degraded inside autolysosomes, steady-state protein levels are routinely monitored as a readout of autophagic flux alongside LC3-II accumulation, and the protein is implicated in Paget disease of bone, amyotrophic lateral sclerosis, and several cancer pathways.
