SMYD1 is a muscle-restricted histone methyltransferase from the SET and MYND domain-containing protein family that catalyzes methylation of histone H3 at lysine 4 and has a well-documented role in cardiac and skeletal muscle differentiation, myofibril assembly, and chromatin remodeling during cardiomyocyte stress responses. This mouse monoclonal primary antibody from Antibodies.com recognizes the SMYD1 target and has been validated for Western blot analysis on rat samples, supporting studies of muscle development, cardiac hypertrophy, and exercise-induced adaptation in rodent models.

The reagent is supplied as 100 µL of purified mouse monoclonal in 0.1 M Tris-Glycine buffer at pH 7.4 with 150 mM NaCl, 0.2% sodium azide, and 50% glycerol as a cryoprotectant, shipped unconjugated so that investigators can pair it with HRP, fluorescent, or biotinylated secondary detection systems of their choosing. Storage at -20 °C in single-use aliquots is recommended to preserve activity and minimize freeze-thaw damage; the sodium azide preservative should be washed from membranes before enhanced chemiluminescence development if azide-sensitive HRP substrates are used downstream.

Reactivity is reported for rat SMYD1, which makes this clone appropriate for Western blot workflows on left ventricular, soleus, and EDL tissue lysates as well as on differentiating C2C12 cultures and primary neonatal cardiomyocytes. Researchers investigating SMYD1-dependent transcriptional programs during heart failure, pathological remodeling, and developmental myogenesis use this reagent to confirm expression changes observed in transcriptomic and ChIP-seq data sets.

For Western blot, typical conditions include 5–20 µg of total protein per lane separated on SDS-PAGE, transferred to PVDF or nitrocellulose, blocked in 5% non-fat milk or BSA, and probed at 1–5 µg/mL of primary antibody followed by an anti-mouse secondary. The unconjugated format also permits custom fluorophore or biotin conjugation for multiplex panels probing additional histone methyltransferases and contractile proteins in parallel.
